Stability and Storage: | Room temperature: paraffin-embedded tissue blocks and slides Refrigerated up to 3 days: blood, bone marrow, fresh tumor or tissue
Processing: | Do not centrifuge. Do not aliquot. Do not freeze. Keep specimens at appropriate storage temperature until ready to transport.
Transportation: | Transport all specimen types at room temperature. Specimens must not freeze in transit. Transport to Pathology Department at Foothills Medical Center.
Method: | PCR followed by agarose gel detection
Method and Interpretation of Results: | DNA is extracted and PCR amplified using primers targeting the major translocation cluster (MTC) of the Bcl-1 locus and the immunoglobulin heavy chain (IgH) gene joining region (JH). Samples are run in parallel with positive, negative, and no template controls. Some breakpoints (as much as 50%) occur at distant loci and will not be identified by this particular test. Samples producing a PCR product in the valid size range are considered positive for the Bcl-1 translocation.
