Stability and Storage: | Room temperature for 24 hours. Refrigerate if storage is prolonged. | ||||||||

Transportation: | Transport specimens at room temperature to Arthur J.E. Child Comprehensive Cancer Centre Flow Cytometry within 24 hours of collection. | ||||||||
Method: | Multiplex PCR followed by capillary electrophoresis. | ||||||||
Method and Interpretation of Results: | Chimerism status is determined by amplification of short tandem tetranucleotide repeats with a panel of fluorochrome labelled primers at 16 chromosomal loci using the AmpFlSTR Identifiler Plus kit from Applied Biosystems (ABI). Amplified products are detected using the SeqStudio genetic analyzer. Pre-transplant recipient and donor allele patterns are compared to post-transplant recipient allele patterns. ABI Genemapper Software is used to determine the relative donor allele percentage.
Test sensitivity for detecting mixed chimerism by this method is routinely 3-5% minor allele as determined using in-house validation with defined mixtures of DNA. | ||||||||
