Method: | Molecular Pathology North PCR followed by single base extension and mass spectrometry Molecular Pathology South (FMC) PCR followed by single base extension and mass spectrometry | ||||||||
Method and Interpretation of Results: | Molecular Pathology North DNA extraction from formalin-fixed paraffin-embedded (FFPE) tissue was performed using the Thermo Fisher MagMAX FFPE DNA/RNA Ultra Kit on the KingFisher Apex purification system. Testing was performed using the Agena iPLEX HS Colon panel and analyzed using an Agena MassArray matrix assisted laser desorption-ionization time-of-flight mass spectrophotometer (MALDI-TOF MS). This multiplex assay is capable of detecting the following mutations: BRAF G469E, D594G, V600E; EGFR S492R, KRAS mutation in codons 12, 13, 59, 61, 146, and K117N, K117R; NRAS mutations in codons 12, 13, 59, 61, 146, and K117N, K117R; PIK3CA E542K, E545K, H1047R, H1047L. BRAF V600K mutations will be reported as V600E by this assay. The estimated sensitivity of this assay is 5%. Molecular Pathology South (FMC) Analysis for BRAF, KRAS, NRAS, EGFR and PIK3CA mutations is performed after PCR amplification using a single base extension technique (Agena iPLEX Colon HS assay) and mass spectrometry on the MassArray instrument. This multiplexed assay is capable of detecting BRAF mutations G469E, D594G, and V600E, KRAS/NRAS mutations at codons 12, 13, 59, 61, 117, and 146, and PIK3CA mutaions at codons 542, 545 and 1047. BRAF V600K mutations will be reported as V600E by this assay. The estimated sensitivity of this assay is 5%. | ||||||||
