Paraffin-Embedded Tissue:

Can be submitted as whole block, precut scrolls, or as blank unstained sections on coated slides.
Representative H&E to accompany the specimen.
Slides to to be reviewed by pathologist, minimum tumour percentage is 10% in area of interest.

Stability and Storage:

Room temperature: paraffin-embedded tissue blocks and slides

Transportation:

Transport all specimen types at room temperature.
Transport to Pathology Department at Foothills Medical Center.

Method:

PCR followed by single base extension and mass spectrometry

Method and Interpretation of Results:

Analysis for CTNNB1, GNAS, KIT, PDGFRA, IDH1, IDH2 mutations is performed after PCR amplification using a single base extension technique (Agena iPLEX Pro chemistry) and mass spectrometry on the MassArray instrument.

The estimated sensitivity of this assay is 10%.
