Specimen Collection Requirements:

Amniotic or Other Fluid

Submit 25.0 – 30.0 mL of fluid in sterile polypropylene centrifuge tube, and
A maternal blood sample must be collected for maternal cell contamination studies

Chorionic Villi

Submit 30.0 mg in sterile polypropylene centrifuge tube containing CVS transport media, and
A maternal blood sample must be collected for maternal cell contamination studies
If submitting to Cytogenetics North: CVS is received Monday to Thursday, 08:00 to 15:00 only. Cannot be collected to be received on Fridays.

Blood – Umbilical Cord

Submit one lavender top EDTA containing 1.0 mL cord blood, and
A maternal blood sample must be collected for maternal cell contamination studies

Blood – Peripheral

Newborn to 12 months: Submit one lavender top EDTA containing 1.0 to 3.0 mL of blood.
12 months to Adult: Submit one lavender top EDTA containing 3.0 to 5.0 mL blood.

Tissue

Submit a minimum of 20.0 mg tissue in a sterile container with NO fixative, and
Preferred specimen: umbilical cord, skin, spleen, lung, thymus, kidney, or muscle.
Other samples are acceptable but quality of DNA obtained may be suboptimal for this assay i.e., products of conception or placental tissue. Placental or products of conception (POCs) are at risk of maternal cell contamination and will be tested as a last resort if they are the only specimen available.
A maternal blood sample must be collected for maternal cell contamination studies

Cytogenetic Analysis, Constitutional Requisition
Please ensure all required information, including patient's clinical history/indication, is provided on the requisition.

Multiple tissue specimens from a patient require a separate requisition and a separate specimen bag for each specimen.

Stability and Storage:

Room temperature up to 24 hours: amniotic or other fluid, chorionic villi
Room temperature up to 7 days: blood
Refrigerated up to 48 hours: fresh tissue

Processing:

Do not centrifuge, aliquot or separate specimens.
Do not put tissue specimens in formalin.
Maintain specimens at appropriate storage temperature until transported.

Transportation:

Specimens should be received in the laboratory on the day of collection (preferred).

Specimens that cannot transported to be received during the Cytogenetics Laboratory hours of operation (Monday through Friday 08:00 to 16:15) should be stored at appropriate temperature and forwarded to the laboratory with the next available courier run.

If specimens are sent from outside of Edmonton, please call the laboratory at 780-407-1542 or fax shipping information to 780-407-3059 to inform the lab of expected arrival times and mode of transport.	If specimens are sent from outside of Calgary, please call the lab at 403-955-7375 or fax shipping information to 403-955-3000 to inform the lab of expected time of arrival and mode of transport.
Amniotic or other fluid, Blood and CVS specimens: Transport at room temperature to Cytogenetics Laboratory via UAH Specimen Control WMC 4B2.10. Amniotic or other fluids received on a Friday after 3:00pm will not get CMA on direct (uncultured specimen); CMA will be performed on cultured cells.	Amniotic or other fluid, Blood and CVS specimens: Transport at room temperature to the Alberta Children’s Hospital Cytogenetics Laboratory B0-150.
Tissue specimens: Transport fresh tissue at refrigerated temperature to Molecular Genetics Laboratory via UAH Specimen Control WMC 4B2.10.	Tissue specimens: Transport fresh placental or fetal material to Anatomical Pathology at the Alberta Children's Hospital in an appropriate sized container. For fetal specimens from outside Calgary, please call ACH Anatomical Pathology for further processing and handling information. Anatomical Pathology will forward all fetal specimens at refrigerated to Cytogenetics Laboratory to ACH Cytogenetics Laboratory B0-154.

Amniotic Fluid:
Any amniotic fluid specimen arriving with a request for only CMA testing, will have pre-analytical Rapid Aneuploidy Detection (RAD) testing added, and possibly reflex Karyotype testing if there is an abnormal RAD result.

Chorionic Villi

Cytogenetics North	Cytogenetics South
The collection of the chorionic villi is only performed at the Maternal Fetal Medicine Clinic, Royal Alexandra Hospital in Edmonton.	The collection of the chorionic villi is only performed at the Maternal Fetal Medicine Center, EFW Radiology in Calgary.

Any CVS sample arriving with a request for only CMA testing, will have pre-analytical Rapid Aneuploidy Detection (RAD) testing as well as Karyotype testing added.

Other information:
RAD testing is performed and reported by the University of Alberta Molecular Genetics Laboratory or by the Alberta Children’s Hospital Cytogenetics Laboratory.
