Clinical Indications: | This screen is a simple and rapid method for the initial investigation for mucopolysaccharidoses (a.k.a., disorders of glycosaminoglycan catabolism). | ||||
Ordering Alert: | This test should not be used to monitor known mucopolysaccharidosis patients on treatment. For monitoring known patients, please order Quantitative Total GAGs (Monitoring) | ||||

Specimen Collection Requirements: | First morning void is preferred. Collect a 5.0 to 10.0 mL random urine specimen. | ||||

Processing: | Specimen can be submitted in primary container or aliquot tube. If specimen is transferred to secondary aliquot tube or container, write "urine" on label. Freeze specimen. | ||||
Transportation: | Transport frozen specimens on dry ice. Samples must not thaw during transport. | ||||
Method: | Spectrophotometric | ||||
Method and Interpretation of Results: | The dimethylmethylene blue (DMMB) dye forms a complex with sulfated GAGs present in urine, which can then be measured spectrophotometrically at 520nm.
Abnormal screen results will be followed up with MPS Investigation, which includes Uronic Acids quantitation and Glycosaminoglycan Electrophoresis, Qualitative. | ||||
