Specimen Requirements: For Constitutional Source: Peripheral or Cord Blood
Newborn to 6 months: 6 months to Adult:
1 - 2 mL blood in a Sodium Heparin tube (dark green, not PST). 3 - 4 mL blood in a Sodium Heparin tube (dark green, not PST).

For Prenatal Source: Amniotic Fluid, Chorionic Villi or other prenatal specimens
Amniotic or Other Fluid: Chorionic Villi:
10 – 20 mL in sterile centrifuge tubes. 10 – 30 mg in sterile container with transport media.
For possible microarray, minimum of 25 mL split into 3 centrifuge tubes. For possible microarray, minimum of 15 mg.

For Tissue Source:
Skin / Tissue Biospy: Buccal Smear:
1 cm3 skin or tissue in a sterile container with tissue transport media. 1 smear from each cheek on a separate glass slide. Buccal smears require a buccal smear collection kit.

Stillborn / POC fetal or placental tissue: refer to Rapid Aneuploidy Detection (RAD), fetal tissue (ACH).
 
Min. Sample Required: If unable to collect the minimum as per specimen requirements, contact the laboratory.
 
Specimen Processing: For Peripheral or Cord Blood specimens
  • DO NOT centrifuge or separate specimen.
  • DO NOT refrigerate or freeze specimen.
  • Store and transport whole blood specimen at room temperature.

    For Amniotic Fluid, Chorionic Villi or other prenatal specimens
  • DO NOT refrigerate or freeze specimen.
  • Store and transport specimens at room temperature.

    For Tissue specimens
  • DO NOT put tissue in formailn.
  • DO NOT freeze.
  • Store and transport at 4 - 8oC.

    Specimen Handling:Transport peripheral blood, cord blood, prenatal specimens or buccal smears at room temperature to the ACH Cytogenetics Laboratory B0-150.
    Buccal smear should be received in lab within 30 minutes of collection.

    Transport skin / tissue biopsy at 4 - 8oC to the ACH Cytogenetics Laboratory B0-150.

    Specimensshould be received during the Cytogenetics Laboratory hours of operation (Monday through Friday 08:00 to 16:15). If transport is delayed, store at appropriate temperature and forward to the laboratory on the next business day.

    Turn Around Times by Specimen Type
    Blood: Routine 4 weeks, Stat 1 week
    Amniotic fluid: 2 weeks
    Chorionic villi: 3 weeks
    Skin / tissue biopsy: 4 weeks
    Buccal smear: 1 week

    Requisition/Form:All specimens must be accompanied by a completed Constitutional Cytogenetics Requisition.
    Ensure all relevant sections of the requisition are complete, including indication of specific FISH probe/test.

    Multiple specimens from a patient require a separate requsition for each specimen.
